Glyceryl Trinitrate is a vasodialator (expands the blood vessels). Glyceryl Trinitrate is used for symptomatic or prophylactic treatment of angina, to treat cardiac failure and during hypotensive or cardiac surgery to prevent myocardial infarction.
Glyceryl Trinitrate also known as Trinitrin, Trinitrin, Trinitrin. . It is of Synthetic origin and belongs to Nitropropane. It belongs to EDRF (NO) agonist pharmacological group on the basis of mechanism of action and also classified in Antianginal Agents and Vasodilators pharmacological group.The Molecular Weight of Glyceryl Trinitrate is 227.10.
Volume of distribution is found to be 3.3 l/kg and plasma protien binding is 60%. Presystemic metabolism is noted to be 75% ±5 and metabolism is reported primarily in Liver. Renal Excretion accounts for partial and plasma half life is 1-3 min.

Angina pectoris, Cardiac failure, Cardiopulmonary oedema, Long-term treatment of healed reflux oesophagitis, Moderate to severe reflux oesophagitis, Myocardial infarction, Oesophageal spasm, Oesophageal spasm, Postoperative hypertension, Prinzmetal's angina, Prophylaxis of angina, Unstable angina.

Interference in Pathology
False decrease in serum cholestrol by Zlotkis-Zak color reaction technique.
Increased urinary conc. of catecholamines and their metabolites.
The severe or irreversible adverse effects of Glyceryl Trinitrate, which give rise to further complications include Tachycardia, Bradycardia, Collapse.
The signs and symptoms that are produced after the acute overdosage of Glyceryl Trinitrate include Hypotension, Flushing, Tachycardia, Coma, Palpitations, Metabolic acidosis, Headache, Cardiovascular collapse, Syncope, Neurologic deficit, Methemoglobinemia, Increased intracranial pressure, Hypoxia, Cyanosis.
The symptomatic adverse reactions produced by Glyceryl Trinitrate are more or less tolerable and if they become severe, they can be treated symptomatically, these include Weakness, Dizziness, Headache, Nausea, Vomiting, Restlessness, Palpitation, Abdominal pain, VertigoX, Urinary incontinence, Syncope, Fecal incontinence, Orthostatic hypotension.

Glyceryl Trinitrate's dosage details are as follows:
0.5 to 1 mg 0.75 (0.75) As recommended. Sublingual As required
glyceryl trinitrate can be repeated every 3 minutes untill pain relieves.
5 to 10 mg 7.5 (7.5) As recommended. Topical glyceryl trinitrate 2% ointment can be used every 4-6 hours as required

High Risk Groups
Drug should not be given to Paediatrics, Pregnant Mothers, patients suffering from Kidney dysfunction, patients suffering from Liver Malfunction, Geriatrics, and Neonates.
If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.
Warning / Precautions
Excessive use of glyceryl trinitrate may cause headache, so reduce the dose & use analgesics (painkiller). Patients with gastrointestinal (GI) hypermotality or malabsorption syndrome should take oral or sublingual sustained release capsules rather than tablets. In terminating treatment of angina, gradually reduced the dose in order to avoid withdrawal reactions. It should not be used in patients with severe hypotension, hypovolaemia marked anemia, heart failure due to obstruction, raised intracranial pressure or angle closure glaucoma. It should be used with caution in patients with impaired kidney or liver function or hypothyroidism.
