Piperaquine Phosphate

Overview

Pipreraquine is 1,3-bis-[4-(7-chloroquinolyl-4)-piperazinyl-1]- propane, is an antimalarial compound belonging to the 4-aminoquinolines. It is a highly liphophilic. Piperaquine Phosphate (PQP) interferes with physiological function of the food vacuole membrane of the trophozoites leading to autophagocytosis of the parasites. It has no marked effect on the ring forms, immature or mature schizonts and the male of female gametocytes.

Pharmacokinetics

Oral absorption of Piperaquine Phosphate is found to be 85% ±5. Volume of distribution is found to be 716 L/kg . Plasma half life is 9.4 days upto 20 days.

Piperaquine Phosphate's dosage details are as follows:

Adult Dosage

16 to 26 mg/Kg21 (21)24 hourlyPOIt is available in combination with dihydroartemisinin. It should be taken with water without food. Doses are given as mg/Kg/day for three days. Maximun 4 tablets each containing 320mg Pipraquine are indicated for upto 100 Kg weight patient. However, there are no data on which to base a dose recommendation in patients weighing >100kg.

Paedriatic Dosage (20kg)
